Things I check:

• Mao's shoulder should be textured
• The big 100 in the middle of the bill should be embossed in exactly the same spot on the reverse side of the bill
• The small 100 near the bottom of the bill should change from gold to green depending on the angle at which you are viewing it

There's also the metal strip running down the middle of the bill (the difference between the real and fake strip in the above picture is pretty clear)

There's also the mao watermark.

Some shops have some kind of UV device that you can stick a bill in to see if it's real.
